Stoke's first leg match with Gillingham was broadcast live on Saturday when Bermudian Kyle Lightbourne scored one goal and had a hand in the other two as Stoke won 3-2.
They will be trying to protect that one-goal lead at Gillingham's Priestfield Stadium, knowing that a draw willbe enough to take them to Wembley for the play-off final on May 27.
Gillingham will be cheered on by a sell-out crowd of 10,000 for the first time this season. The game is also being screened live back to Stoke's Brittania Stadium where 7,500 fans have paid 5 each to watch the match.
International interest in the game also spans to Iceland as the club is under Icelandic ownership. A television audience of around 250,000 watched the first match live in Iceland.
Gillingham, who recently beat Stoke 3-0 at home, are confident of clinching the tie as they bid to reach the Second Division play-off finals for the second straight season. Last year, Shaun Goater's Manchester City scored two late goals to beat them and clinch the final promotion spot to the First Division.
Stoke have a full squad to choose from, with Lightbourne and top scorer Peter Thorne leading their attack, while Gillingham manager Peter Taylor has written off striker Carl Asaba's chances of being fit enough.
Asaba missed the first leg on Saturday with knee ligament damage and now Taylor's best hope is that the 27-year-old will be fit if the Gills reach Wembley.
The winner of the Gillingham-Stoke match will meet either Millwall or Wigan in the final. They played to a 0-0 draw in the first leg.
